Joker truly does not know what he is talking about. Seconds later, he says he was only young when he did that.
Joker claims ignorance. He never learns Batman is Bruce Wayne. He believes he killed Bruce Wayne when he shot him earlier.
He cannot know he was young because he does not know when Batman's parents died. He only knows this happened before the raid on Axis Chemicals.
